Does VS200 slide scanner software align images from different layers automatically?

Yes, there is a tool for automatic image alignment. If you have two layers with one common channel (e.g., DAPI), simply click on the relevant menu item and the tool will do the alignment for you. You also have the option to export data as separate layers to Olympus’ cellSens™ software or other image analysis software.

If you are looking at 8 or 12 different markers, do you still image them in sets of 4?

Yes, when increasing the number of markers, you continue to image them 4 at a time. This way, you can keep using the 4 classical fluorophores: FITC, TRITC, Cy5 and Cy7. This approach avoids bleed-through or crosstalk between fluorophores.

Can modern slide scanning be performed on standard glass slides?

Yes, you can use normal glass slides. There are no special sample preparation requirements—the sample just needs to be paraffin-embedded.

How do you reduce tissue autofluorescence and other noise during image acquisition?

We have tested the Ultivue stains and found that they have a very high signal-to-noise ratio and dynamic range, which reduces the effect of autofluorescence. Also, Ultivue stains do not use a secondary antibody, so you avoid background signal due to nonspecific binding. Finally, choosing the right filters helps to remove background signal.

What is the Virtual Z feature in the VS200 slide scanner?

This is the system’s Z-stack feature, which enables you to do volumetric (3D) imaging by combining multiple exposures at different focal planes. The VS200 scanner also has extended focus imaging (EFI), which can flatten this Z-stack into a single 2D image. This feature is particularly useful when you need a fully focused image from a specimen that is not completely flat.

Does the VS200 scanner have barcode recognition?

Yes, the VS200 slide scanner has an integrated barcode reader that helps you simplify data management. The reader can scan a large selection of 1D and 2D barcode types to capture and record the information automatically.
